Join us for this webinar with Eurofins as we discuss the assessment of potential cardiotoxicity of compounds with Comprehensive in vitro Proarrhythmia Assay (CiPA), the new cardiac safety testing paradigm that includes in vitro assays using human induced pluripotent stem cell derived cardiomyocytes (hiPSC-CM).
 

To adopt this paradigm, we developed a high-throughput hiPSC-CM assay using calcium-sensitive dyes that exhibit fast kinetic fluorescence detected by the high-speed fluorescence imaging platform, the FLIPR® Penta system.

[NEW] Application Spotlight

Multi-parametric assessment of compound-induced pro-arrhythmic effects in human iPSC-derived cardiomyocytes
 
Development of biologically relevant, predictive cell-based assays for compound screening and toxicity assessment is a major challenge in drug discovery. The focus of this study was to establish cardiotoxicity assays compatible with high-throughput screening using human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes.

iPSC-derived cardiomyocytes generate spontaneous, synchronized calcium oscillations. We used high speed fluorescence imaging on the FLIPR® Penta High-Throughput Cellular Screening System to measure the patterns and frequencies of the Ca2+ oscillations in cardiomyocytes as monitored by changes in intracellular Ca2+ levels with the EarlyTox™ Cardiotoxicity Kit. Plus, we show enhanced peak characterization with new ScreenWorks® Peak Pro 2 software.
