- New ABF 2.0 file format
* Requires Digidata 1550 Data Acquisition System
Existing functionality includes the ability to automate the execution of protocols with Sequencing Keys, a variety of recording modes from Gap-Free to Episodic Waveform stimulation, online filtering and leak subtraction, as well as online statistics and support of Automatic Quick Graphs.
The AxoScope 10 Software provides a convenient way to produce background recordings with the included MiniDigi 2-Channel Digitizer. You can monitor cells during inter-sweep periods, or create an overview of the entire day's activities including voice tags. The Clampex and Axoscope applications can acquire data concurrently on the same computer if both a MiniDigi Digitizer and a Digidata System are connected.
The Clampfit 10 Data Analysis Module is a powerful solution for analyzing, graphing, and formatting of all of your Clampex and AxoScope data. The Clampfit Module includes an extensive array of filtering and fitting routines. Functionality includes I-V graphs, power spectrums, and special "linked data views" for threshold (AP), template (minis), and single-channel modes of event detection and analysis.

Amplifiers
Analog signals from many other sources, such as transducers of physical parameters like pressure or light, can be acquired along with or instead of electrophysiological signals. In combination with the Digidata 1550 Low-Noise Data Acquisition System, analog and digital outputs can be used for sophisticated control of equipment in creative experiments.
Software Compatibility
Developer Information
Software libraries are available for programmers who want to write programs that read or write the Axon instruments data file formats produced by pCLAMP 10 (Clampex and AxoScope) data acquisition programs, and are still available for previous versions of the pCLAMP (Clampex and Fetchex), AxoScope, and AxoTape data acquisition programs.
Please note that technical support is not available for writing programs using these libraries.
For pCLAMP 10 (ABF 2):
- File Support Pack 2 for Windows (release build) [ZIP] allows Windows programs to support the ABF and ATF data file formats for AxoScope 1 to 10, Clampex 6 to 10, Fetchex 6, and AxoTape 2, via Windows DLLs. (This build corrects the library in the previous 3/12/07 zip file.)
- File Support Pack 2 for Windows (debug build) [ZIP] is the same as above, but includes extra error trapping code.
- ABF 2 Docs [ZIP] file, provides details of the ABF 2 file header for non-Windows OS support. (Additional information on header details has been added.)
For previous versions of pCLAMP (ABF 1):
- File Support Pack 1 for Windows [ZIP] allows Windows programs to support the ABF and ATF data file formats for AxoScope 1 to 9.2, Clampex 6 to 9.2, Fetchex 6, and AxoTape 2, via Windows DLLs or header offsets.
- File Support Pack for DOS [EXE] allows DOS programs to support the ABF and ATF data file formats for Clampex 6, Fetchex 6, and AxoTape 2, and the EVL events list format for Fetchan 6 and pSTAT 6.
pCLAMP™ 10 Electrophysiology Data Acquisition and Analysis Software is suitable for a wide variety of applications, ranging from electrophysiology to general physiology, from individual cells to networked cells, and from low-noise patch clamping to high-speed sodium channels. Here are some examples of typical applications:
- Action Potentials (APs): Typically recorded from a single cell either extracellularly, or intracellularly in whole-cell current clamp or with sharp electrodes.
- Bilayer Recordings: Measurement of single-channel currents through ion channel molecules inserted in a planar lipid membrane.
- Current Clamp: The amplifier controls the current through the membrane of a patch-clamped cell, while voltage fluctuations, such as action potential, are recorded.
- Electrocardiography (ECG)*: Extracellular population recordings of cardiac activity.
- Electrochemistry (Voltammetry / Amperometry): Electrochemical measurements that allow the detection of substances though oxidation or reduction; typically used for the detection of neurotransmitters in brain slices.
- Electroencephalogram (EEG)*: Extracellular population recordings of brain activity.
- Electromyography (EMG)*: Extracellular population recordings from skeletal muscles.
- Electroretinography (ERG)*: Extracellular population recordings from the retina.
- Excitatory and Inhibitory Post-synaptic Currents (EPSCs and IPSCs): Voltage clamp recordings from a post-synaptic cell while the pre-synaptic cell is stimulated.
- Excitatory and Inhibitory Post-synaptic Potentials (EPSPs and IPSPs): Current clamp recordings from a post-synaptic cell while the pre-synaptic cell is stimulated.
- Long-term Potentiation and Depression (LTP and LTD): Modulation of synaptic transmission in response to preceding stimulation.
- Miniature Post-synaptic Potentials (Minis): Small post-synaptic potentials, typically associated with the release of a single transmitter vesicle.
- Perforated Patch Clamp: Measurement of ionic currents through the membrane of a patch-clamped cell after inserting pore-forming agents in the membrane.
- Single-channel Recordings: The activity of individual ion channels in the membrane of a patch-clamped cell, typically recorded in excised patches.
- Spike Trains: Sequences of action potentials.
- Synaptic Network Recordings: Activity of a network of neurons forming synapses with each other, either in culture or in tissue slices
- Tissue Slices: Recordings from cells still embedded in tissue sections to keep intercellular connections intact; typically whole-cell or perforated patch clamp recordings.
- Voltage-clamp: The amplifier controls the membrane potential of a patch-clamped cell, while ionic currents through the membrane are recorded.
- Whole-cell Patch Clamp: Measurement of ionic currents through the membrane of a patch-clamped cell after rupturing the membrane patch spanning the pipette orifice.
