We will describe methods and present specific binding data for characterization of agonists and antagonists to several GPCR receptors including Mu Opioid, Dopamine D3, and Glucagon GLP1 receptors. We will also describe and demonstrate performance of several other functional assays using HTRF® technology: cAMP detection, p-ERK and p-AKT kinase assays. Studies will focusing on ligand binding and numerous functionality readouts using the same assay platform.
The Tag-lite® cellular platform is ideal to screen cell surface receptor targets, in particular GPCRs. It provides a wide range of assay formats for the investigation of receptor ligand binding and function. The Paradigm plate reader platform with a HTRF® cartridge provides fast, sensitive readout of the binding assay. The combination of the SpectraMax Paradigm® plate reader with the HTRF® and Tag-lite® suite of assays is shown to be a powerful assay platform for cell surface binding and functional assays and well suited for high throughput screening.
